Cell populations transduced with a pooled CRISPR library are emulsified into single-cell droplets with overlap extension RT-PCR reagents. Following thermal lysis and reverse transcription, the overlap extension PCR physically links gRNA transcripts to target cDNAs or antibody-conjugated oligo barcodes from the single cells. These linked amplicons are subsequently analyzed via short-read sequencing to assign multi-omic phenotypes to specific genetic perturbations. b, Generation of the CD81 CRISPRi validation model.
