S2255) and 0.8 % Astra Blue (Santa Cruz Biochem, cat. sc-214558A) for 3 min. Safranin-O preferentially stains lignified cell walls (e.g., xylem vessels) red, while Astra Blue stains cellulose-rich tissues blue, enabling the distinction of vascular and storage tissues.
