Figure Legend Snippet: The co-culture of fibroblasts and monocytes was treated with SB-505124 (5 µM), tofacitinib (1 µM), or IKK-16 (1,5 mM) either immediately (0h) after hydrogels generation or 24h later and subsequent contraction at 72h was compared with vehicle-treated (DSMO) (control). ( A ) Hydrogels treated immediately (0h) with SB-505124 displayed impaired contraction, which did not occur when the inhibitor was added later (at 24h). Inversely, hydrogels receiving tofacitinib at 0h showed strong spontaneous contraction, which did not happen upon treatment after 24h. Blocking the NF-κB pathway did not affect (myo)fibroblast contraction at any time point. Each shape represents the average of 3 technical replicates of one PBMC donor, n = 5. Groups were statistically compared with one-way ANOVA followed by Tukey’s multiple comparisons test (*p<0.05, **p<0.01).
