Article Title: Assays for ubiquitin-like protein ligation and proteasome function in archaea
Journal: Methods in enzymology
doi: 10.1016/bs.mie.2018.12.036
Figure Legend Snippet: Ubiquitin-like protein (Ubl) ligation and proteasome systems of Archaea. The Ubl protein (blue) is activated at its C-terminus by an E1-like enzyme (red). Adenylation (Ubl~AMP) and thioester (Ubl~E1) intermediates are formed. The Ubl is transferred from the Ubl~E1 to the target protein (green) by a mechanism that is stimulated by MsrA. The Ubl is linked by an amide (isopeptide) bond to the target protein. JAMM1 can cleave the Ubl-modified protein at the isopeptide linkage. The activity of JAMM2, AAA ATPases (Cdc48 and PAN), and 20S core particles is required for turnover of Ubl-modified proteins. X?, additional factors may be required.
Techniques Used: Ubiquitin Proteomics, Ligation, Modification, Activity Assay
