Effects of <t>PSB12379</t> (selective CD73 inhibitor) on the metabolism of N 6 -etheno-AMP (eAMP) to N 6 -etheno-ADO (eADO) by sEV isolated from plasma of healthy donors (HD sEV; left panel ) and patients with head and neck cancer (HNC sEV; middle panel ) or melanoma (melanoma sEV; right panel ). sEV samples were incubated for 20min with eAMP (100 µmol/l) in the absence (control) or presence of PSB12379 (40 µmol/l). After 20min incubation, the concentration of eADO was measured in each sample using high pressure liquid chromatography with fluorescence detection. In the presence of PSB12379 , the levels of eADO were below the detection limit (<dl) of our assay system (1 pmol injected on column). Results are shown in bar graphs with individual data points, means and SEMs.
