( A ) Cell lines expressing Trop2, namely, MDA-MB-468 and letrozole-resistant MCF-7 (LTLT-Ca), were checked for Trop2 expression by Western blot. Expression was compared relative to the control cells HEK293t, which do not express Trop2. ( B ) Respective cell lines were treated with either HEK293T E or NB60 E for 48 h. After EVs treatment, protein lysates were collected and ran for Western blot. Trop2 knockdown was compared to the control, the cells not treated with the respective EVs. ( C ) LTLT-Ca cells were seeded onto a 96-well plate and were treated for 48 h with varying concentrations of the respective EVs. After treatment of cells with the indicated concentrations of EVs, MTT assay was performed to determine the viability of the cells. ( D ) LTLT-Ca cells were seeded into 100 mm tissue culture dishes and were treated for 48 h with either Sacituzumab (Sac) or NB60 E. After treatment, the cells were collected, and protein lysates were prepared for Western blot. Trop2 expression was probed to compare knockdown of the said antibodies in LTLT-Ca cells.
